Assay #1: Determined by its ability to induce apoptosis in HeLa cells. The expected ED50 for this effect is 5.0–10.0 ng/ml. Assay #2: The ED50 was determined by a cytotoxicity assay using HT-29 cells is 0.05 ng/ml, corresponding to a specific activity of 2×107 units/mg.

Reconstitution

Centrifuge the vial prior to opening. Reconstitute in water to a concentration of 1.0 mg/ml. Do not vortex. This solution can be stored at 2–8°C for up to 1 week. For extended storage, it is recommended to further dilute in a buffer containing a carrier protein (example 0.1% BSA) and store in working aliquots at –20°C to –80°C.

Summary

IFNγ is a homodimeric glycoprotein containing approximately 21 to 24 kD subunits. Production of IFNg is a function of T cells and NK cells. IFNγ is produced during infection by T cells of the cytotoxic/sup­pressor phenotype (CD8) and by a subtype of helper T cells, the Th1 cells. IFNγ is a diagnostic tool for distinguishing tuberculous from other nontuberculous ascites. It is important in the design and monitoring of immunotherapy for multiple sclerosis.
