Description

IL-12 is a potent regulator of cell-mediated immune responses and it induces IFN-γ production by NK and T cells. It is produced by activated monocytes/macrop­hage cells, B lymphocytes and connective tissue-type mast cells. Among its biological activities, IL-12 promotes the growth and activity of activated NK, CD4+ and CD8+ cells, and induces the development of IFN-γ-producing Th1 cells. Recombinant Murine IL-12 is a 75.0 kDa heterodimeric glycoprotein consisting of disulfide-linked 35 kDa (p35) and 40 kDa (p40) subunits (506 total amino acid residues).

Biological Activity

Assay #1: The ED50 was determined by the stimulation of IFN-gamma production by murine splenocytes co-stimulated with IL-18 is < 0.1 ng/ml, corresponding to a specific activity of > 1×107 units/mg. Assay #2: Determined by its ability to stimulate the proliferation of 2D6 cells. The expected ED50 is ≤0.1ng/ml, corresponding to a specific activity of ≥1×107 units/mg.

Reconstitution

Centrifuge the vial prior to opening. Reconstitute in 1× PBS, pH 7.2–7.4 to a concentration of 0.1–1.0 mg/ml. Do not vortex. This solution can be stored at 2–8°C for up to 1 week. For extended storage, it is recommended to further dilute in a buffer containing a carrier protein (example 0.1% BSA) and store in working aliquots at –20°C to –80°C.
