PubMed Product Details
Technical Data

Type

Sandwich ELISA, AChE-labelled antibody

Applications

Plasma-EDTA+PHMB

Sample Requirements

20 µl/well

Shipping

On ice. Upon receipt, store the product at the temperature recommended below.

Storage/Expiration

Store the kit at -20°C. Under these conditions, the kit is stable until expiration date (see label on the box).

Calibration Range

250 - 2 pg/ml

Limit of Detection

The limit of detection, calculated as the concentration of Acylated Ghrelin corresponding to the NSB average (n = 8) plus three standard deviations is 0.8 pg/mL. Due to the minimal plasma dilution (1:5), the limit of detection in the samples is less than 5 pg/mL.

Intra-assay (Within-Run)

CV = 6,8% (20h at +4°C) CV = 5.7% (3h at RT)

Inter-assay (Run-to-Run)

CV = 7 % (20h at +4°C) CV = 6.1% (3h at RT)

Spiking Recovery

99.5% (20h at +4°C) 98.6% (3h at RT)

Dilution Linearity

105 %

Summary

Ghrelin, an endogenous ligand for the growth hormone secretagogue receptor, is synthesized principally in the stomach. It stimulates food intake and transduces signals to hypothalamic regulatory nuclei that controls energy homeostasis. The peptide consists of 28 amino acids, with a n-octanoylation of the serine-3 residue, which is necessary for the biological activity mentioned below. Ghrelin is present in the peripheral circulation under two forms: acylated and unacylated.
